Penrose was born to James Doyle Penrose, a successful portrait painter, and Elizabeth Josephine Peckover, daughter of Lord Peckover, a wealthy Quaker banker. He was raised in a strict Quaker family and was brother to Lionel Penrose, a noted medical geneticist.

His contributions to psychiatry and genetics, including the development of intelligence tests and the Penrose method for global voting systems, left a lasting impact on medical genetics and public health.
